I can't answer, but I can speculate:
i. In the main game, probably not, because I don't think they'd want to say, "We're out of vowels," when the puzzle has just started.
ii. In the bonus puzzle, I could easily see having a puzzle where the only vowel sound comes from a Y:
_RY (a lot of these)
STY
FLY
WHYS
TRYST
CYST
MYTH
(There's also a Welsh word CWM, but I can't imagine them using it.)

It's possible though unlikely that they used any of the following:
MMMM MMMM MMMM MMMM (song title, 1994)
M & Q (fictional characters, from the Bond films)
Protecting idiots from themselves just leads to more idiots.
